Opening her acceptance speech with "I'm 160lbs and can catch a dick whenever I want," the New Yorker went on to keep the audience fully entertained.
We weren't the only ones who loved Amy's hilarious speech - some of the guests immediately took to Twitter to show their love...
Here's our best bits about Amy's speech...
Referring to former winners claiming they aren't glamourous enough to be at the event: I'm NOT nervous. I'm very glamorous. I don't feel out of place here.
On winning her award: It will be very exciting when what I am doing isn't trailblazing anymore…but I'm very happy to have gotten this.
On Goldie Hawn: I'm SO desperate for a selfie with Goldie Hawn that I'm not even going to ask. I would just want it for me, like I would just look at it every night and be like "Goodnight".
On women: I just think women are BADASSES.
She told us on the red carpet: "Women are bad asses and owning it more."
When asked her advice for younger women: Definitely get vaccinated for HPV.
On shaving: My mom told me I didn't need to shave above my legs above the knee. I basically like Christopher Robin down there.
ON, erm, we're not sure: Tonight, I had one goal. To take my underwear off at the end of the night and not make it look like I blew my nose on it.
When asked where she would take GLAMOUR.com News Editor Leanne Bayley on a night out, she responded: We would probably be at my apartment and we would probably just be drunk. Okay Amy - you're on.
British actress and comediene Jennifer Saunders presented the award after telling us how it was her daughter who had alerted Saunders to Amy's talent.
Inside Amy Shumer has already seen her nominated for a Primetime Emmy Award, Critics Choice Television Award and win the prestigious Peabody Award this year. The 33-year-old can now proudly add her GLAMOUR award top the mix.
